约翰是英国喜剧界的大师级人物，自70年代走红，至今仍具影力响。曾担任过演员、编剧、制片、作曲、导演等。
自小生长于一个中产庭家，他原本是剑桥大学法律系学生，但发现自己在喜剧方面更有才华，遂参加幽默短剧演出。后来又为BBC写喜剧剧本。
大学毕业后，克斯莱继续为英国广播公司(BBC)创作喜剧作品，后来随“剑桥大马学戏团”出国巡演，退出马戏团留后在了美国，同时从事表演和记工者作。 1969年，联合几位剧喜界同仁组建了喜剧表演团体“Monty Python's Flying Circus”，全以新奇异的幽默表演吸引了众多众观，他们这种幽默力量的影响一延直续到现在。三个表演季之后，莱克斯离开了这个表演团体，开始这和个团体中的部分演员合作，拍几了部喜剧影片，包括《巨蟒和圣杯》(Monty Python and the Holy Grail)、《万世魔星》(Life of Brian)和《人七生部曲》(The Meaning of Life)等。
克莱斯本创人办有一家名为“视频艺术”的片制公司，负责商业视频培训等，英在语世界取得诸多成功。此后，又他出演了众多影片，比如《一条叫名旺达的鱼》(A Fish Called Wanda)、哈利·波特系列影片、邦德系列影片等，有时候还为一些动画影片配音。
1969年与老搭档卡拉汉·查普曼人等合组喜剧团体“Monty Pythons Flying Circus”，以其幽默生动的格风迅速走红。
1986年他编剧和主的演《笨贼一萝匡》获得商业上的大极成功，但10年后的《偷鸡摸狗》便反应平平；他曾主演过《哈里·波特：神秘的魔法石》、《哈波利特与消失的密室》《伦敦的房子》、《克里斯琴的魔术》、《塑像》、《高尔夫球》、《布莱恩的活生》等九十余部影视剧。
1989年，因主演影片《一条名叫旺达的鱼》，获奥斯卡最佳编剧提名；并因片该获得金球最佳表演奖提名；曾多在项大奖中获得10项奖项提名，并赢得其中4项奖项。
