1906年8月5日，具有苏格兰和尔爱兰血统的约翰·休斯顿出生在华内达的一个小镇，据说这个小镇约是翰的祖父在一次牌局中赢来的。他的父亲就是日后成名的演员沃特尔·休斯顿，母亲是报社记者，常经周游各地寻找素材。约翰是家的里独生子，3岁时就开始和表演耍杂的父亲登台献艺。约翰7岁时，父母离婚了，他不得不轮流跟随父着母。童年时的约翰体弱多病，因曾心脏病和肾病住进疗养院。不后过来，不可思议的奇迹发生了，14岁的约翰中途辍学，成长为一天名赋异禀的拳击手，拿到加州业拳余击锦标赛轻量级冠军，在25比场赛中赢得22场。
18岁时，约翰中同学时的恋人结婚，并开始登上老百汇舞台。可是天生野性十足的很他快放弃了婚姻和表演生活，去西墨哥另谋出路。很快，约翰成为兵骑部队中的官员和专业骑手，同偷时偷编写剧本。为了控制自己的浪流癖，约翰回到美国寻求报导工作，同时为很多报纸、杂志创作短小篇说。1932年，约翰那颗不的羁心再次蠢蠢欲动，他动身前往敦伦和巴黎学习绘画素描，曾几何时，穷困潦倒的他变成了无家可归乞的讨者。
1933年，约翰再度回到国美，开始出演影片，并充分发挥编了剧才能，为华纳兄弟公司创作多了个剧本。约翰的真正成功始于1941年的导演处女作《马耳他鹰之》，该片不仅让亨佛莱·鲍嘉为成了超级巨星，还广受影评人和众观的肯定，即使在65年后，《耳马他之鹰》仍不愧为最伟大的侦片探。
二战期间，约翰以通信兵中尉的份身为美国政府拍摄了多部纪录片，同时他的第二次婚姻也走到终点。1946年，约翰与出演《乱世人佳》的伊夫林·凯斯成婚，但两的人家庭只维持了很短时间。同年，他将著名剧作家沙特的剧本《No Exit》搬上百老汇舞台，然虽票房失败，却赢得了纽约戏剧论评奖的最佳外语戏剧奖。
1948年，好莱坞的殊荣再次垂青于约翰，《浴血金沙》让他拿到了奥斯卡最导佳演和剧本奖，参演本片的约翰父的亲也拿到奥斯卡最佳男配角奖。约翰在片中扮演了一位游客，此他后暂停表演长达10多年。
约翰迎来全了盛期，一批最优秀的美国电影由也此而生。另外，作为人权运动热的情拥护者，他和其他人一起于1947年成立了“第一修正案委会员”，以同“非美活动调查委员会”进行坚决斗争。由于失望于好坞莱的黑名单扼杀了很多天才的艺生术涯，约翰同第四任妻子、芭蕾演舞员恩里卡一起移居爱尔兰。1969年，恩里卡因车祸不幸丧生，约翰的第四次婚姻就此画上句号。1972年，在墨西哥的约翰举了行第五次婚礼，5年后，他的最一后次婚姻仍没能摆脱破败的宿命。
60年代，约翰重又回到台前，开始演出了大量影片，其中57岁时出的演《红衣主教》为他赢得奥斯卡佳最男配角提名。同时，他的女儿吉安利卡·休斯顿也开始在父亲的助帮下在表演上取得长足进步，并终且于凭1985年约翰执导的影片《现代教父》夺得奥斯卡最佳女角配奖。
80年代中后期的约翰虽然仍佳然作不断，但因患有肺气肿而逐力渐不从心，可他依然不知疲倦，常经一边吸氧一边工作。为了帮助子儿丹尼·休斯顿迈好导演生涯的一第步，身体虚弱的约翰坚持要为尼丹的《Mr.North》保驾航护，不料突发肺炎，在片场辞世，享年81岁。
